Both the show and the anime are aligned with everything. so def recommend the anime. But if you feel impatient waiting for the next episodes to release or feel it's going too slow then you're free to pick up the manga if you wanna know what happens or go at your own pace. I Also recommend reading through the official fan book if ya wanna get a good idea of everyone;s timeline in the series.

As Darya and Yakov take the perilous journey to sea to seek medicine for Connie but will die at sea sometime in July...
Connie dies soon later in the same month... with Shamil contracting the illness as well and decease in August

There's no what if if youre already living it. Petrification day was June 3, 2019 after all. Tsukasa and Gen would never meet the Trio. Senku would likely be working at J.A.X.A. or N.A.S.A either going to the moon or pursuing the same thing as Dr. Xeno, possibly work beside Xeno as a Colleague.
Taiju and Yuzuriha would be happily married and settled down with kids. As Taiju would likely get into something like Construction (Since he's muscley) And Yuzuriha be running a fabric/craft store.
Tsukasa would continue his career in the MMA.
Gen would still be a show man.

Yes, but not through legal means, prolly domestic partners. Where Kohaku is the Dominant and prolly be forceful. Get that Senku isnt too into it, but maybe Kohaku will idk light a fire under him to get him moving. Guy needs a little push if you know what Im saying.
